I brought a new 750gb sata drive and I want to do a full copy of my 250gb drive. I want it to include everything like the mbr, music, games, programs and work.

What would be the easiest way to do this so that I can erase my 250gb drive to use as portable storage.

There are many ways to do that. Windows has its own backup program. Or you can use Acronis True Image.
